Agenda One is a brilliant tool, bit the problem I expierience is following:
Any new calender entry or deleting a calender entry takes a loooong time until it becomes effective (> 1 minute). New tasks or contacts appear immediately on the calendar screen.

What device are you using? Did you soft reset your device to see if that clears the issue? We have not yet had other users report that specifically adding or deleting calendar entries causes this type of delay and we are so far not able to duplicate the problem on various devices. Please also check that you have sufficient available memory on the device to operate the software. Check also to see that you do not have a number of other programs running at the same time in the background which can cause performance issues with any software.

I am using a HTC Touch Pro and there shoulsd be enough memory left (195 mb for data). Also I did a warmstart after installing agenda one.
Would agenda one have a problem with many calandar entries? I have more than 5000 going back 7 years.

Yes, that much data will be a problem for most calendar software. The program must read so much data before displaying a view or result. You may need to consider changing the setting for the amount of data that you send to your device. Are you aware that in ActiveSync when you double clik 'Calendar' that you have a number of options to choose the time frame of data that is sent to the device from your PC. You will still have all the old history on your PC, but sync only a portion of it to the device.
